The aims of the project are (1) Characterization of new effects of cyanobacteria on important biomarkers of tumor promotion; (2) Assessment of biochemical and cellular mechanisms; (3) Characterization of new bioactive compounds; (4) New information for risk assessment of toxic cyanobacteria.
The hypothesis of the project assumes the production of unidentified cyanobacterial metabolites with tumor promoting potencies. Our studies employ the rat liver stem-like WBF344 cells (an established model for studies of tumor promoters). We investigate the effects of cyanobacterial samples on GJIC, activations of mitogen-activated protein kinases ERK1/2 and cellular proliferation. Using fractionation followed by HPLC and MALDI-TOF MS we will characterize physico-chemical properties of bioactive metabolites.
